If I transfer my US number to Mint Mobile for 2FA purposes, can I use that at the same time as I use a local SIM to access data etc in destination country?
Or can you only access US # separately from local data?

You can, but with the changes Mint made to their international roaming charges this week its no longer the most cost-effective option.
Go for Tello or US Mobile instead and save yourself $10 a month
